{"id":2037,"date":"2020-04-25T09:30:30","date_gmt":"2020-04-25T07:30:30","guid":{"rendered":"https:\/\/www.umrs1176.universite-paris-saclay.fr\/?page_id=2037"},"modified":"2025-09-01T17:13:04","modified_gmt":"2025-09-01T15:13:04","slug":"themes","status":"publish","type":"page","link":"https:\/\/www.umrs1176.universite-paris-saclay.fr\/index.php\/themes\/","title":{"rendered":"Th\u00e8mes"},"content":{"rendered":"<p data-start=\"77\" data-end=\"508\">L\u2019unit\u00e9 INSERM U1176 \u00e9tudie tous les aspects de la r\u00e9action h\u00e9mostatique, qu\u2019ils soient <span style=\"color: #0000ff;\">physiologiques <span style=\"color: #ff0000;\"><span style=\"color: #000000;\">ou<\/span> pathologiques<\/span><\/span>, ainsi qu\u2019aux troubles associ\u00e9s r\u00e9sultant d\u2019une r\u00e9ponse inappropri\u00e9e. Notre force et notre originalit\u00e9 r\u00e9sident dans <span style=\"color: #0000ff;\">une approche int\u00e9grative<\/span> qui combine l\u2019expertise sur les <span style=\"color: #ff9900;\">acteurs cellulaires de l\u2019h\u00e9mostase (les plaquettes)<\/span> et sur ses <span style=\"color: #ff9900;\">acteurs mol\u00e9culaires (facteurs pro- et anticoagulants).<\/span><\/p>\n<p data-start=\"510\" data-end=\"1158\">Le syst\u00e8me h\u00e9mostatique des mammif\u00e8res est en effet le fruit d\u2019une \u00e9volution extr\u00eamement sophistiqu\u00e9e, ayant conduit \u00e0 l\u2019\u00e9laboration d\u2019une cascade de complexes (pro)enzymatiques destin\u00e9s \u00e0 produire une r\u00e9ponse rapide en cas de l\u00e9sion vasculaire, en parall\u00e8le avec le d\u00e9veloppement d\u2019un syst\u00e8me cellulaire sp\u00e9cialis\u00e9 et d\u00e9di\u00e9 : les plaquettes. Ces deux syst\u00e8mes se sont dot\u00e9s de m\u00e9canismes de contr\u00f4le sp\u00e9cifiques permettant d\u2019\u00e9viter une r\u00e9activit\u00e9 excessive. L\u2019int\u00e9grit\u00e9 de ce syst\u00e8me est essentielle au bon d\u00e9roulement de l\u2019angiogen\u00e8se, du d\u00e9veloppement et de la r\u00e9paration tissulaire dans des conditions physiologiques.<\/p>\n<p data-start=\"1160\" data-end=\"1392\">En revanche, lorsque l\u2019\u00e9quilibre de l\u2019hom\u00e9ostasie est rompu, le syst\u00e8me h\u00e9mostatique peut \u00eatre d\u00e9tourn\u00e9 et contribuer \u00e0 des processus pathologiques tels que <span style=\"color: #ff0000;\">l\u2019h\u00e9morragie, la thrombose, l\u2019inflammation et\/ou la progression tumorale.<\/span><\/p>\n<p data-start=\"1394\" data-end=\"1919\">D\u2019un point de vue m\u00e9canistique fondamental, la complexit\u00e9 des interactions entre le contenu (le sang) et le contenant (les vaisseaux) reste encore loin d\u2019\u00eatre totalement \u00e9lucid\u00e9e. Il est cependant \u00e9vident qu\u2019un d\u00e9faut de communication entre les principaux acteurs peut jouer un r\u00f4le cl\u00e9 dans la pathogen\u00e8se des troubles de la coagulation, des maladies (ath\u00e9ro)thrombotiques et de l\u2019inflammation. Cette connaissance est d\u2019une importance capitale pour le diagnostic et le d\u00e9veloppement de nouvelles strat\u00e9gies th\u00e9rapeutiques.<\/p>\n<p style=\"text-align: center;\"><span style=\"color: #0000ff;\"><strong>TROIS THEMES SCIENTIFIQUES SONT ABORDES DANS L&rsquo;UNITE U1176\u00a0\u00a0<\/strong><\/span><\/p>\n<p style=\"text-align: justify;\"><div class=\"su-accordion su-u-trim\"> <div class=\"su-spoiler su-spoiler-style-fancy su-spoiler-icon-plus su-spoiler-closed\" data-scroll-offset=\"0\" data-anchor-in-url=\"no\"><div class=\"su-spoiler-title\" tabindex=\"0\" role=\"button\"><span class=\"su-spoiler-icon\"><\/span>Plaquettes : Aspects cellulaires\/vasculaires<\/div><div class=\"su-spoiler-content su-u-clearfix su-u-trim\">\n<p style=\"text-align: justify;\">R\u00e9f\u00e9rent :\u00a0<a href=\"https:\/\/www.umrs1176.universite-paris-saclay.fr\/index.php\/organigramme\/\" target=\"_blank\">Fr\u00e9d\u00e9ric Adam<\/a><\/p>\n<p style=\"text-align: justify;\">Le th\u00e8me plaquettes\/aspects cellulaires et vasculaires a pour but de d\u00e9crypter les m\u00e9canismes cellulaires et mol\u00e9culaires, aussi bien fondamentaux que pathologiques, r\u00e9gulant <strong>la quantit\u00e9 de plaquettes et leurs fonctions h\u00e9mostatiques<\/strong>. Les plaquettes jouent un r\u00f4le cl\u00e9 dans l\u2019arr\u00eat du saignement, les recherches au sein de l\u2019unit\u00e9 INSERM abordent les diff\u00e9rents versant de l\u2019activation plaquettaire aussi bien les phases pr\u00e9coces que les phases plus avanc\u00e9es\u00a0comme la formation du thrombus (figure ci-dessous).<\/p>\n<p style=\"text-align: justify;\"><a href=\"https:\/\/www.umrs1176.universite-paris-saclay.fr\/wp-content\/uploads\/2020\/04\/Screen-Shot-2020-04-21-at-16.05.50.png\"><img loading=\"lazy\" decoding=\"async\" class=\"aligncenter wp-image-2076 \" src=\"https:\/\/www.umrs1176.universite-paris-saclay.fr\/wp-content\/uploads\/2020\/04\/Screen-Shot-2020-04-21-at-16.05.50.png\" alt=\"Screen Shot 2020-04-21 at 16.05.50\" width=\"675\" height=\"266\" srcset=\"https:\/\/www.umrs1176.universite-paris-saclay.fr\/wp-content\/uploads\/2020\/04\/Screen-Shot-2020-04-21-at-16.05.50.png 964w, https:\/\/www.umrs1176.universite-paris-saclay.fr\/wp-content\/uploads\/2020\/04\/Screen-Shot-2020-04-21-at-16.05.50-300x118.png 300w\" sizes=\"auto, (max-width: 675px) 100vw, 675px\" \/><\/a>Un des axes de recherche porte sur l\u2019implication du <strong>cytosquelette, des prot\u00e9ines de signalisation et de la r\u00e9gulation de la signalisation calcique, <\/strong>(notamment les pompes Serca3), dans la formation et l\u2019activation des plaquettes. Le cytosquelette est un acteur essentiel par son implication sur leur formation, la morphologie des plaquettes, la s\u00e9cr\u00e9tion,&#8230; Mieux comprendre, mais \u00e9galement d\u00e9finir de nouveaux acteurs impliqu\u00e9s dans ces m\u00e9canismes, sont des enjeux essentiels notamment dans des pathologies induisant des d\u00e9fauts morphologiques (macro- ou microplaquettes) ou encore du nombre de plaquettes (thrombop\u00e9nie) ou bien des d\u00e9fauts d&rsquo;activation plaquettaire (thrombopathies). Le laboratoire\u00a0utilise aussi bien des m\u00e9gacaryocytes en culture que des plaquettes, humaines et murines et des lign\u00e9es cellulaires.<\/p>\n<p style=\"text-align: justify;\"><a href=\"https:\/\/www.umrs1176.universite-paris-saclay.fr\/wp-content\/uploads\/2020\/04\/Screen-Shot-2020-04-21-at-16.02.47.png\"><img loading=\"lazy\" decoding=\"async\" class=\"aligncenter wp-image-2074 \" src=\"https:\/\/www.umrs1176.universite-paris-saclay.fr\/wp-content\/uploads\/2020\/04\/Screen-Shot-2020-04-21-at-16.02.47.png\" alt=\"Screen Shot 2020-04-21 at 16.02.47\" width=\"725\" height=\"368\" srcset=\"https:\/\/www.umrs1176.universite-paris-saclay.fr\/wp-content\/uploads\/2020\/04\/Screen-Shot-2020-04-21-at-16.02.47.png 991w, https:\/\/www.umrs1176.universite-paris-saclay.fr\/wp-content\/uploads\/2020\/04\/Screen-Shot-2020-04-21-at-16.02.47-300x152.png 300w\" sizes=\"auto, (max-width: 725px) 100vw, 725px\" \/><\/a>Un second axe de recherche porte sur la r\u00e9gulation de la quantit\u00e9 de plaquettes en p\u00e9riph\u00e9rie. Dans cette th\u00e9matique, notre laboratoire, s\u2019int\u00e9resse tout particuli\u00e8rement au<strong> r\u00f4le de l\u2019acide sialique<\/strong> et l\u2019exposition du \u03b2-galactose, un signal de s\u00e9nescence r\u00e9gulant la clairance des plaquettes.<\/p>\n<p style=\"text-align: justify;\">Enfin, un axe important de nos recherches porte sur l\u2019\u00e9tude de pathologies plaquettes connues telles que les <strong>filaminopathies A,<\/strong> <strong>la maladie de Willebrand de type 2B <\/strong>ou encore l\u2019\u00e9tude de pathologies associ\u00e9es \u00e0 des \u00e9v\u00e8nements <strong>h\u00e9morragiques inexpliqu\u00e9s<\/strong>. Dans ce cadre, notre objectif est d\u2019\u00e9valuer et de mieux comprendre les m\u00e9canismes mol\u00e9culaires sous-jacents \u00e0 ces pathologiques. Un second volet de cet axe porte sur le suivi de patients trait\u00e9s lors de protocoles cliniques (suivi des patients Wiskott-Aldrich trait\u00e9s par th\u00e9rapie g\u00e9nique).<\/p>\n<p style=\"text-align: justify;\">Pour mener \u00e0 bien l\u2019ensemble de ces \u00e9tudes, nous disposons de mod\u00e8les multiples, tels que :<\/p>\n<p style=\"text-align: justify;\">&#8211; mod\u00e8les animaux,<\/p>\n<p style=\"text-align: justify;\">&#8211; acc\u00e8s \u00e0 des \u00e9chantillons de patients,<\/p>\n<p style=\"text-align: justify;\">&#8211; lign\u00e9es cellulaires primaires et d\u00e9velopp\u00e9es au laboratoire,<\/p>\n<p style=\"text-align: justify;\">&#8211; notre laboratoire travaille en \u00e9troite collaboration avec l\u2019h\u00f4pital et la recherche justify;\">Les projets de recherche s\u2019inscrivant dans ce th\u00e8me s\u2019articulent autour de la cascade de la coagulation (voir sch\u00e9ma ci-dessous) dont le but principal est l\u2019arr\u00eat du saignement lors d\u2019une br\u00e8che vasculaire.\u00a0Les mol\u00e9cules pro-coagulantes sont ici repr\u00e9sent\u00e9es en vert.<\/p>\n<p style=\"text-align: justify;\"><a href=\"https:\/\/www.umrs1176.universite-paris-saclay.fr\/wp-content\/uploads\/2020\/04\/coagulation-cascade1.jpg\"><img loading=\"lazy\" decoding=\"async\" class=\"aligncenter wp-image-2053 style=\"text-align: justify;\">Nous nous int\u00e9ressons particuli\u00e8rement \u00e0 l\u2019\u00e9tude des<strong> relations structure-fonctions<\/strong> des prot\u00e9ines de l\u2019h\u00e9mostase en nous focalisant sur celles qui sont absentes ou mut\u00e9es dans des pathologies h\u00e9morragiques constitutionnelles. C\u2019est le cas du<strong> facteur Willebrand<\/strong> dans la maladie de Willebrand, <strong>le facteur VIII dans l\u2019h\u00e9mophilie<\/strong> <strong>A<\/strong> ou l<strong>e facteur IX dans l\u2019h\u00e9mophilie B<\/strong>. Notre int\u00e9r\u00eat s\u2019\u00e9tend \u00e9galement aux prot\u00e9ines qui peuvent \u00eatre utilis\u00e9es en th\u00e9rapeutique pour traiter ces pathologies, comme le <strong>facteur X<\/strong> ou le <strong>facteur VIIa<\/strong>.<\/p>\n<p style=\"text-align: justify;\">Afin d\u2019\u00e9tudier ces prot\u00e9ines, nous utilisons un certain nombre d\u2019outils biologiques dont des <strong>nanobodies ou VHH <\/strong>(mini-anticorps d\u00e9velopp\u00e9s par les cam\u00e9lid\u00e9s) que nous d\u00e9veloppons au sein du laboratoire. Susen (CHU Lille). Ce projet vise \u00e0 mieux comprendre la <strong>maladie de Willebrand acquise<\/strong> d\u00e9velopp\u00e9e par les patients plac\u00e9s sous assistance circulatoire m\u00e9canique (ou souffrant de st\u00e9nose aortique) et \u00e0 r\u00e9pondre au challenge th\u00e9rapeutique pos\u00e9 par les saignements (principalement gastro-intestinaux) observ\u00e9s chez ces patients. Nous nous int\u00e9ressons non seulement \u00e0 l\u2019effet purement anti-coagulant de ces diff\u00e9rentes mol\u00e9cules mais aussi \u00e0 leurs effets cellulaires qui leur permettent d\u2019\u00eatre impliqu\u00e9s dans divers processus au-del\u00e0 de l\u2019h\u00e9mostase et en particulier l\u2019inflammation. Nos projets actuels sur les Serpines visent d\u00e9sormais 1) \u00e0 <strong>cibler l\u2019antithrombine<\/strong> dans un but de r\u00e9-\u00e9quilibrer l\u2019h\u00e9mostase dans l\u2019h\u00e9mophilie et 2) \u00e0 mieux comprendre les <strong>relations structure-fonctions de l\u2019inhibiteur d\u00e9pendant de la prot\u00e9ine Z, le ZPI <\/strong>qui est une des serpines les moins bien caract\u00e9ris\u00e9es jusqu\u2019\u00e0 pr\u00e9sent. L\u2019objectif est \u00e9galement de mieux comprendre le r\u00f4le du ZPI dans des pathologies inflammatoires et thrombotiques, mais \u00e9galement dans l\u2019h\u00e9mophilie. Pour cela, nous nous appuyons sur le d\u00e9veloppement au sein de notre groupe de <strong>nanobodies ou VHH<\/strong> (mini-anticorps d\u00e9velopp\u00e9s par les cam\u00e9lid\u00e9s) dirig\u00e9s contre cette prot\u00e9ine.<\/p>\n<p style=\"text-align: justify;\">Concernant le syst\u00e8me de la prot\u00e9ine C, notre focus porte principalement sur la <strong>prot\u00e9ine S<\/strong> et nous souhaitons \u00e9valuer s\u2019il est possible de moduler l\u2019activit\u00e9 anticoagulante de la prot\u00e9ine S en utilisant des nanobodies dirig\u00e9s contre cette prot\u00e9ine.
